Frylock is the best character and is really the glue that keeps the show together. Shake is the funniest character however. So I have to go with Meatwad, he's right in the middle.

My favorite is Frylock. He brings the most to the table for me, perhaps because he seems to be the most sane one or at least acts that way most of the time. Meatwad would be next, for the randomness and somewhat innocence of him. Shake goes last because he just seems to do the same schtick.
